Semaglutide is a GLP-1 receptor agonist that slows gastric emptying, lowers appetite signaling, and improves insulin sensitivity. The injection (Wegovy) was approved in 2021 and produced the largest weight loss of any GLP-1 trialed up to that point. The oral pill formulation uses an absorption enhancer (SNAC) to get the molecule across the gut wall — making it the first GLP-1 weight-loss medication that doesn't require an injection.
OASIS-4 evaluated high-dose oral semaglutide in adults with obesity or overweight with weight-related conditions, without diabetes. Patients on the top oral maintenance dose lost a mean 14 to 16 percent of body weight over 64 weeks compared with a small change on placebo. The trial established oral semaglutide as a viable alternative to injection for chronic weight management.
Wegovy Pill (oral semaglutide) is a prescription medicine. Important safety information applies. The most common side effects are gastrointestinal, usually mild during titration. There is a boxed warning for thyroid C-cell tumors, based on rodent studies.
