Leqvio® is a small interfering RNA (siRNA) therapy that “silences” PCSK9 in the liver. With more LDL receptors active, cholesterol is cleared from the blood more effectively. In large clinical trials, LDL dropped by ~50% and ApoB by ~40–50%, even on top of statins or ezetimibe.123
After the first two doses (month 0 and month 3), you only need one injection every 6 months. This steady dosing avoids the daily burden of pills or frequent antibody injections, helping more patients stay on track long term.45
By cutting LDL and ApoB in half, Leqvio® reduces key drivers of plaque buildup in arteries. Pooled data from large trials suggest a ~24–26% reduction in heart attacks and strokes. Larger outcome studies are ongoing.678
Leqvio® is generally well tolerated. The most common side effect is mild injection site reactions. Serious events, including liver or kidney issues and new diabetes, occur at similar rates to placebo. Four-year follow-up shows no new safety concerns.39

Using RNA interference (RNAi), inclisiran pairs with the messenger RNA (mRNA) that encodes PCSK9 and marks it for breakdown. Less mRNA means less PCSK9 protein.
PCSK9 normally flags LDL receptors for destruction. With PCSK9 reduced, more receptors survive, stay active, and keep pulling LDL cholesterol out of the bloodstream.
Bottom line: this removes the brake on your body’s natural LDL-clearing system.
